Darius Demetrius Slay was born on January 1, 1991, in Brunswick, Georgia. He attended Brunswick High School, where he played football as a defensive back and wide receiver. Slay was a two-way player in high school, and he earned all-state honors as a senior after catching 29 passes for 676 yards and eight touchdowns.
After high school, Slay attended Itawamba Community College in Fulton, Mississippi, where he played football for two seasons. As a sophomore, he was named a first-team All-American by the National Junior College Athletic Association (NJCAA) after recording 42 tackles, six interceptions, and 14 passes defensed.
Slay then transferred to Mississippi State University, where he played for the Bulldogs football team from 2011 to 2012.
